Introduction to pH Probes
pH regulation is closely linked to mitochondrial bioenergetics, ATP synthesis, proton transport, organelle function, metabolic reprogramming, and cell survival. In mitochondria, matrix pH and proton gradients are directly related to oxidative phosphorylation and energy conversion. In other subcellular compartments, pH changes can reflect lysosomal function, endosomal trafficking, autophagy, apoptosis, inflammation, and tumor microenvironment acidification.
pH probes can be designed using multiple sensing principles, including fluorescence intensity change, ratiometric fluorescence, near-infrared signal switching, genetically encoded pH sensors, optical fiber sensing, electrochemical ISFET sensing, MOF-based luminescent sensing, and microfluidic pH detection.
For mitochondrial and cellular applications, pH probe development requires careful optimization of pKa, dynamic range, response time, reversibility, photostability, cytotoxicity, subcellular localization, anti-interference performance, and compatibility with fluorescence microscopy, confocal imaging, flow cytometry, plate-reader assays, high-content imaging, microfluidic platforms, and selected environmental or industrial testing workflows.
